As far as I know, chloroquine may accumulate in the lysosome and gradually increase lysosomal pH and impair its function. Therefore, I first thought that chloroquine-treated cells stained with. CQ is a weak base and therefore it can raise the pH of cellular compartments. This has led to the assumption that CQ blocks the autophagic flux through the same mechanism as BafA 1, which increases lysosomal pH and thus inhibits the activity of resident hydrolases 14 – 16. Rlyso, a highly selective and sensitive pH sensor, can stain lysosomes with a novel lysosome-locating group, methylcarbitol. Rlyso was successfully used to detect lysosomal pH changes during apoptosis or induced by chloroquine while avoiding the “alkalizing effect” on lysosomes of current lysosomal probes with nitrogen-containing sidechains.

  4. Chloroquine overdose is a life-threatening emergency and should be managed with cardio-respiratory and hemodynamic support, monitoring of potassium along with management of arrhythmias and convulsions, as necessary. A patient who survives the acute phase and is asymptomatic should be closely observed until all clinical features of toxicity resolve.

Chloroquine raises the pH in host-cell lysosomes, which interferes with viruses’ attempts to acidify the lysosomes, a prerequisite to formation of the autophagosomes that cells use to eat themselves. In the Cell Research paper, the researchers found that the drug was effective at inhibiting the virus as it was both entering and exiting cells. It accumulates in the acidic lysosome as it becomes protonated, increasing the pH of the lysosome 35,36. Following exposure to concanamycin A, BMPR-II accumulates in the lysosome, but with chloroquine treatment BMPR-II appeared to be increased at the plasma membrane in addition to a degree of lysosomal accumulation. Chloroquine is a lysosomotropic weak base, which in the monoprotonated form diffuses into the lysosome, where it becomes diprotonated and becomes trapped. Protonated chloroquine then changes the lysosomal pH, thereby inhibiting autophagic degradation in the lysosomes.

What next if plaquenil doesn't work? Mar 24, 2011 Had headaches with the plaquenil so went to the sulfar and predisone. Sulfar was a waste of time and since I had a little help from the plaquenil he switched me back to that with pain meds for the headaches and body. Recently i went back for app. and plaquenil had stopped doing anything so he added the methotrexate to it. I did not want to take it.

Do either hydroxychol/Plaquenil put you at risk for greater.
